After two days of deliberations the league appeals court also confirmed the five-year ban given Genoa president Enrico Preziosi by the disciplinary commission last week.
Genoa was found guilty of sporting fraud for fixing its final match last season against Venezia. Genoa won 3-2 and was promoted from Serie B to Italy's top league after a 10-year absence.
Genoa was also docked three points from its tally next season.
Some supporters of Italy's oldest club had threatened violence if Genoa's relegation was confirmed. Fans forced the referee to abandon Sunday's first-round Italian Cup match with Catanzaro by throwing lit flares and firecrackers onto the field.
The league didn't say which team would replace Genoa in Serie A.
